The effects of blocking sigma receptors are something I've wanted to explore for a long time. The only antagonist I know of is iboga, but that interacts with so many other receptors its impossible to know what part of its effects are due to sigma antagonism. I was recently prescribed amitryptiline, I thought it would be a reliable sleep aid I could use temporarily, but it only works for one day, then it has the opposite effect, it keeps me awake at night. I noticed this trend happen with trazodone and mirtazapine but it took a few weeks for them to stop working on me, so my guess is that amitryptiline is doesn't work on me due to my previous use of mirtazapine. I don't know what all this means pharmacologically, but I just noticed amitryptiline is a sigma agonist so that caught my attention.
