ADX-47273 is an mGluR5 positive allosteric modulator. It shows pro-cognitive and antipsychotic effects in animal studies.

 
It has a half-life around 2h in rodents. I don't know how bioavailable it is orally.
 
It has not been tested in humans. In animal studies, it has been used at doses ranging from 1 to 300 mg/kg, with pro-cognitive effects at 1-10mg and antipsychotic effects at 10-100mg. No harmful effects were noted except for catalepsy at 300mg/kg.
 
DISCLAIMER: I haven't gone through the literature carefully, I just looked at the article I linked and another review article. So don't assume everything I write is precisely true, like those doses for example, I might have them not accurately.
 
Anyway. I might get it for myself. I'm interested in mGluR modulation.
 
Anybody else interested?
 
Also I'm wondering something. It hasn't been tested in humans. It looks very safe, but humans are no rodents. I know people have here tested drugs that have never been tested in humans before. What kind of things should I take into consideration? Are there some risks that typical rodent studies easily miss?
 
I don't expect anyone to have clear answers for me. It's a rather unknown substance and nobody can tell me if it's safe. All I'm asking is general thoughts and discussion on the topic of taking drugs that have not been tested in humans.

I'm seriously considering testing this on myself...

 

As this compound has not been tested for any other routes of administration than intraperitoneal injection (maybe some other injection too, I haven't checked), it's not known how this would behave if administered orally.

 

I'm considering injection (IV, IM or SC), but I'd rather use something else, as I have never injected anything before. What do you think would be the most reliable way to get the drug to the brain: intranasal, sublingual or rectal? Obviously we can't know what works and what doesn't for 100% certainty, it would have to be investigated. But I'm talking about odds. What's the most likely successful route of administration?
