Positive results with fasoracetam for ear ringing (tinnitus)

I started taking fasoracetam for its (supposedly) great nootropic effects and noticed none of that at a low dose of 20-25mg. However, I did notice some mild sedation (GABA-B pathway) and less ear ringing. This is the first thing that has that effect on my ear ringing, not counting a certain 5HT2A agonist that made it worse.

I did some searches and there is a connection between GABA-B and tinnitus and the fasoracetam effects were reported before: https://www.tinnitus...soracetam.8941/

 

While this is anecdotal, we may have a winner here.

 

To be fair, I have a mild case of tinnitus. On good days it's soft hissing, on bad days it's high pitch tone. Noticeable, but not disabling yet. However, I have a family history of old age hearing loss (presbycusis) and that goes together with tinnitus.

Did nothing for me though. Lysine tablets work for me a bit, they are anxiety-reducing so that might be the mechanism of action.

weird, i have been taking Lysine in various doses for a few months and noticed no impact on tinnitus.

Shows how much of a pain this affliction is really. Not one tinnitus seems the same. I had mine since I was 18, no clear indication of how/why I developed it. Fucking tinnitus  :ph34r:

Fasoracetam does not bind to gaba-b.

Glutamate agonists upregulate gaba receptors while gaba agonists upregulate glutamate receptors.

Fasoracetam's effect on gaba-b is supposed to occur after its binding has ceased.

 

If gaba-b receptor activation works for tinnitus then it is almost certainly because gaba-b counteracts glutamate. Overexcitation is linked to causing tinnitus via loss of nerve cells and also I believe short-team independently of loss of nerve cells.

It doesn't make sense to me why fasoracetam would work against tinnitus immediately when you take it. In fact it caused tinnitus for me once (very high pitched sound).

 

Note that fasoracetam by binding to mglur2/3 is antipsychotic on this disease model of hallucinations. People with schizophrenia, schizotypalpd etc., often experience tinnitus, hissing or a high pitched sound. That's not a hallucination but it co-occurs with, precedes or proceeds hallucinations or pseudohallucinations.

Update on my "experiment."

i woke up early AM and immediately noticed tinnitus, very noticeable, 3/10, similar to last PM. i remembered about Fasoracetam, took 25mg only with water and went back to sleep. i woke up 1 hr later, well rested and noticed tinnitus was present, 2/10, but didn't bother me at all. i got a cup of coffee and started a weekend project. 1 hr later, 2 hrs after Fasoracetam, there was no tinnitus (0/10) and then i took rest of my stack, ate some, and went back to work.

Now, it's mid afternoon and there is minimal hissing, 1/10. The tinnitus will probable came back later in PM as it has been doing for the last 3 days, except for the first day, when i took Fasoracetam twice a day and it covered the entire day (but had problems going to sleep).

i did things differently today and results are inconclusive. I didn't take fasoracetam this AM, instead i took my regular stack. tinnitus was mild the whole morning, 1-2/10. I took about 20mg fasoracetam at noon, and now, 2.5 hrs later there is still some mild tinnitus, 1/10. at least it didn't get worse.

Jack Black are you not reading? How the hell would glutamate stimulation help tinnitus? Tinnitus is primarily caused by excessive glutamate. Fasoracetam is also known to CAUSE tinnitus as it did in me.

well, it's known to you only, but somehow it didn't CAUSE tinnitus in my case.

furthermore, today, the first day without fasoracetam, tinnitus is worse.

we just went through this with NAC in another thread. activation of mGluR2/3 INHIBITS NMDA signaling, duh.

I can again confirm that fasoracetam causes tinnitus. I took fasoracetam, had no tinnitus before and then immediately right after taking it I had low-level tinnitus.
