I also take a glass of grapefruit juice on the first day of my rapa cycle to keep the level high for 2 or 3 days and then I let the Rif remove the remainder quickly.

I take probiotics the day after I take the Rifa, but yes that is a concern.

 

 

the microbiome is literally thousands of different organisms. a probiotic wont just magically make it right

 

ABX fuck up your microbiome, taking ABX voluntarily when you dont even have a dangerous infection is, impo, reckless and just really really not a good thing to do. 

 

In my opinion this protocol will likely SHORTEN your life, not lengthen it.

I've been trying the same thing. At the moment 8mg of Rapa once a week. About 150mg of rifa once a day but I sometimes skip. I also take about 1000mg of allantoin spread throughout the day. I made the mistake at first of taking 1000mg of metformin with about 500mg of synthetic allantoin... but I've now quit the metformin.


How did you decide on a gram of allantoin per day? That seems potentially high. Your rapa and rifa look like they are at pretty motivated levels, as well.

Rapa is the standard amount recommended by Dr Green for males. The dosage of allantoin is about the same as the old dosage of metformin I used to take (they are similar compounds). The experiments with the worms used the ratio of 1:5 between rif and allantoin. So I basically just followed it and started on 150 - 200mg of rif, which is lower than the standard amount used. I intend to cycle the rif (one month on, one month off)

I've changed my rifa to 150mg every Tuesday, Thursday and Saturday morning. I take half a glass of grapefruit juice on Saturday as well to counteract the excessive breakdown of rapa. I take my 8mg Rapa every Sunday evening with another half glassof grapefruit to keep the concentration high before the rifa starts breaking it down. Because of its low absorbtion through the digestive system I've left the allantoin about the same at 1gm daily.

8 mg rapamycin with grapefruit juice? If I took that my mouth sores would be unbearable. How old are you? Even Dr. Green, who is his low 70's, takes 6 mg a week and does not recommend grapefruit juice. I have talked to him personally.

Grapefruit doesn't increase the concentration in the blood, all it does is stop the body breaking down the rapa for about 2 to 3 days. Rifa has the opposite effect of breaking down the rapa twice as fast. They counteract each other. It is just a matter of getting the balance right.

I seem to feel a little down and my Libido seems low when I am on the rifa. That's the main reason I cut the dosage back. As an aside, I have also noticed that allantoin doesn't seem to cut sugar cravings quite as well as metformin.

that's very interesting, I did not know that.

I'm not a chemist, so where can I learn about their similarities as it pertains to human metabolism?

 

Also, regarding allantoin, according to wiki, some gut bacteria will likely degrade unabsorbed allantoin to ammonia -- could this be the reason for rifampin (other than its well-known property as  inducer of the hepatic cytochrome P450 enzyme system). Of course, there are better antibiotics for this purpose. Besides, you take allantoin daily, while rifa once a week, no?

 

I think degradation of allantoin to ammoina by gut bacteria is a concertning issue. How do you take allanoin to improve its absorption? I see solubility in water is 0.57 g/100 mL, which is not bad at all. Do you wash it it down with a glass of water or simply take with food?

 

I'm asking, coz, though I'm not planning on doing rifa, I find allantoin very interesting. I may consider taking it even though, despite it being known and used (externally) for a long, long time, there is not much on ingesting it by humans (though surely it has been tried). I find your experience very interesting.

oh! I found this exhaustive review on safety of allantoin, including its ingestion by humans, if anyone is interested:

Most mammals process purines to allantoin... However, higher apes and humans do not possess urate oxidase, the final step in the process; thus, purines in humans become uric acid, which is excreted in urine.

I am using Comfree, which is a product by Solaray. It’s benign comfrey root, which contains an unknown amount of an evidently “safe” dose or Allantoin. I am in contact with the company and they are trying to find out how much Allantoin is in the product. Not sure if they will succeed. I came up with the dose and schedule for Rifampcin with Dr. Alan Green, whom I’m sure you are familar with if you know Blogskenny.

 

 Thanks for doing your research and sharing. I trust the good doctor Green. I imagine Rifampicin is dosed by weight. I am a very small person, any idea what the dose per kg or lb would be?

 

BTW,m Doesn't doctor Green Rx metformin for his Rapa patients? I'm curious to know why he doesn't have the same takeaway from the research that you do. Have you discussed it? I'm not sure it's an exact comparison, but I took berberine instead of metformin. The latter is supposedly the synthetic or synthetically produced version of the former. I felt awful on it. It wasn't even that I had any expected GI symptoms, it just made me feel like a zombie at 500mg BID and even 500mg SID was more than my body could make sense of. I threw out the bottle because I never want to make the mistake of taking it again. Do you (or others) have any idea if I should expect the same from metformin?

Do you know his recommended dosage for women? I think I have it in an email but can't find it. I stopped the Rapa last year when I couldn't tolerate the berberine. I'm glad to read that the metformin/berberine type supplements/drugs are not necessary during Rapa trials. I am looking for an ideal dose of Rapa for 40-something women. If you konw what the Dr Recommends, please share.  Thank you!
