Has anyone tried all three substances 5-HTP, Tryptophan and Sam-e separately? How do they compare?

From what I've read, it may depend on your metabolism as to which is better. I have tried all three and I generally stick with 5htp. I liked SAMe but the price is steep and the mood boosting benefits seemed to level off.Tryptophan works fine as well, but 5htp works even better for me and is generally cheaper.

Which is better from a side effects point of view?

5-htp is not safe, especially its combination with vitamine b6. Side effects similar to ssri drugs are possible.

Tryptophan is precursor for 5-htp and all depends on the steps before converting to 5-htp, so it's safer at least.

SAME is complex but at least it helps to create tetrahydrobiopterin.

And I wouldn't combine them.

I've never experienced negative side effects from any of these substances.

I gained a lot of weight on Tryptophan I mean a huge amount even when I was eating my regular portions of food. How much Tryptophan did you take?

In my experience with all three they do different things.  Same helps with joint pain and energy levels.  Tryptophan helps with sleep.  5-htp I've had the least success with but is reputed to help more with mood.

I find Same the most "powerful" and that it plays well with Modafinil.

iirc bacopa also acts as a weak SDRI, also boosting gaba + glutamate slightly

Bacopa monniera leaf extract up-regulates tryptophan hydroxylase (TPH2) and serotonin transporter (SERT) expression: implications in memory formation.

Lithium modulates tryptophan hydroxylase 2 gene expression and serotonin release in primary cultures of serotonergic raphe neurons.
[Regulatory effects of Wuzhuyutang (Evodiae prescription) and its consisting herbs on TPH2 promoter].
