I respond very well to NMDA enhancers/glutathione enhancers (sarcosine, NAC, pregnenolone) and respond *horribly* to PQQ. I heard that it effects glutathione synthesis in some way, which probably effects the redox aspect of NMDA receptors. For me, it gave me significant brain fog about an hour after ingesting, without fail, no matter what other things I had been taking. So if you have hypoglutamatergic function(or suspect you do), be careful with this one.

@Luxflux: I am very interested in your response to PQQ and how you've tied that to NMDA function. I also have had really awful experiences with PQQ, very shortly after taking it. I have taken two different brands, 2 small doses and one half-dose on three separate occasions without changing anything else and taken separate from any other supplements.

I would say "brain fog" was part of it, but the experience was more like something I imagine diabetics in severe hypoglycemia experience: Couldn't think/concentrate, irritable, weak/fatigued, super-depressed or "down" mood. The worst of this lasted for hours and didn't really get better for another day. About an hour or two after taking, each time, there was also this unusual craving for carbs and sweets, which is not at all normal for me.

Until I saw your post, all my research just turned up that PQQ gave results for everyone falling on spectrum somewhere between "wonderful and energy-boosting" and "did nothing." So, I had been thinking perhaps it was a clue to some metabolic weirdness with me.

How did you arrive at glutamatergic hypofunction as the cause? Interestingly enough, I am beginning to (cautiously) experiment with pregnenolone with very positive results. NAC is also a staple of mine.
I wish I could say it way a scientific conclusion, but it's mostly just by correlating the results of PQQ ingestion with other substances I've taken that alter Glutathione/Glutamate functioning in the brain. For instance, vinpocetine antagonizes glutamatergic function, as does ashwaganda. When I tried both of these supplements, they effected me like PQQ did. I am not versed in chemistry, but I do know that PQQ has *some* effect on the redox sites involving Glutathione/Glutamate/NMDA. What that effect is I don't know. I can also say cautiously that I don't understand the chemistry behind anything I take, so it is possible that the information I offer is partially or totally innaccurate (thought I'm not just making it up =P ).

Beyond that, I take CoQ10 and Lipoic acid(which as far as I know effect mitochondrial function, among other things) with only positive results. In light of these two pieces of information, I concluded as best I could that it was probably due to the relationship with the glutamate system.

Short of brain fog, I don't remember specific symptoms. I may or may not have experienced the things you are saying. I still have some PQQ left, I may try one capsule this weekend and see what happens. If I do I'll report back.

Regardless of the actual causes behind my poor experiences, at least there is an n = 1 case where someone with low NMDA function had a bad experience with PQQ. Hope this helps you find some answers.
That's very interesting. I got brain fog and sluggishness from trials of ashwaganda as well... but I don't recall those being as profoundly unpleasant as PQQ. Have not tried Vinpocetine. ALA is a daily staple that I feel is required to keep my attention/energy levels up.

This may be a segue to a new thread or PM, but have you ever taken any of the commercial genetics tests (e.g., 23andMe?) I have a small number of fairly rare results for which it would be interesting to see if any match up. My curiousity is particularly focused on some SNPs on the CRF, HPD (homozygous for "Hawkinsinuria,") also CYP2D6*6.

Any family history of schizophrenia? I believe glutamatergic hypofunction is a part of one of the models for underlying causes. I have close family members on both sides with SCZ.

 

 

I was diagnosed as schizophrenic at age 13 (I'm 32 now). The prognosis for teenage onset is rather poor, but I was fortunate to beat the odds. I've not taken medication for 10 years now, and my IQ has increased about 15 points over the last 20 years (It's supposed to degenerate over time, a full standard deviation is quite uncommon). Initially I remedied it via high dose EPA fish oil, but found that compromised glutathione/NMDA systems were probably to blame.

 

I have not taken any genetic testing, but from anecdotal evidence from family members I believe one of the genetic sources to my illness is problems with calcium ion channels. Do you have any snps that relate to that? IMO, this is one of the cores to schizophrenia and related illnesses, though there are probably many paths to the same pathology.

 

I did take another pqq cap, problems were still there, but they were more mild. I think it is due to the increased antioxidants I am taking that cross the BBB (longvida and NA-R-ALA in particular).

I have FMS/CFS and started taking PQQ recently and it has made a huge difference in my mental health as well as fatigue. Nothing has ever made any difference to the crippling fatigue I experience day in day out, so I was so happy to have come across this. I also started taking ubiquinol (Coq10 did nothing for fatigue) which also has the same effect as the PQQ (or maybe it is boosting what the PQQ has regenerated?) but sometimes when I take them together I become really angry and can end up in an uncontrollable rage. B12 also does this to me so maybe it is a methylation issue.

That's great! How long did you take it? And how much?

Did you go the hypothyroid route of www.stopthethyroidmaddness.com?

I have CFS as well and did a lot already and have quite a recovery already but the last bits are the toughest. I notice nothing from 20 mg PQQ (sublingual or oral) but I am only for a week on it. I also take resveratrol for mitochondrial biogenesis and R-ALA to clear up mitochondria. If successful I am going to add quercetin which can cross BBB and promote biogenesis there.

Is the 52nd item if tried since 2003.

I have Severe Allergies since 1958. 62 years old. SMOG changed things a lot for me. Allergies to pollen's and mold's lasted 4-5 months at that time.

Now 11 Months!

 

I Juiced Quite a bit from 1999-2003. I used a L'equip 110.5 at that time. After 2 weeks of juicing I felt like I could lift

a Building!! I Juiced 2500+ 12 oz. glasses of Carrot-Parsley-Apple-Celery& Spinach.

I don't care what type of juicer you have use it.

 

I won't go into more details as to the many items I've tried right now.

PQQ is the subject.

 

I was at yahoo news when an ad for pqq  came up.

Did a lot of research--Wiki--YouTube as well.

I decided to go local.

 

They had Jarrow BioPQQ I took my 1st cap on 8-6-15 @ 11am.

I took the 2nd 12 hours later. This is a 20mg.

First day

Light pain at bridge of nose.

Head- Brain Light pain. Left arm .

Just all over Healing Crisis.

 

I took the Jarrow for 4 Days and took  1/2 day of Monday OFF!!

My order from amazon--Health thru Nutrition 20mg 120 ct.- got here in 2 days.

I started it late Monday. HTN also has 108mg of Calcium.

I took a HTN at 6am Today 8-12-15.

I started Juicing again 1-20-15. I think I'm at appox. 361 glasses. 13oz. avg.

I usually Juice --sandwich-Juice. My Juice is now Apple-Carrot-Celery.

 

I haven't had Healing Effects like this since  1994.

H2o2 started then and continues.It doesn't taste good but it is effective.

Haven't used it in 60 days.

 

I've also been taking Omega oils since 2005.

ProCaps DHA really helped with the Jarrow PQQ.

 

I will continue with PQQ.

I think it is Very Beneficial.

Memory Loss better.

Well

After 10 days I will say this PQQ is something Special. Let's say by chance this is the "Silver-Bullet"

Are Health Food Stores in my area Afraid of it ?

How many people buy Herbal Products like most other's buy the same meds. month after month year after year.

 

I went to 3 HFS in the last 5 days. 2 of the Busiest stores had 1 Brand of PQQ.The slow store had 3.

Today the Busy store was Wiped-Out of their regular products.The Family is Really Educated in the Biz.

This article says PQQ "has to be bound to glycine to penetrate and exert this effect in whole brain." How would one go about binding PQQ to glycine?

3. PQQ stimulates glycine modulatory sites by indirect antagonizing via removing glycine from the synaptic cleft.

4. PQQ binding the unsaturated sites with D-serine in the brain stimulates glycine modulatory sites.

Digging up old threads is fun. :P

My friend took it and stopped after a month. No lasting effects longer than 2 weeks. During that month he reported better energy levels.

I have been taking BioPQQ and I feel more energy and stack it with CoQ10.    I get like an alert buzzed headache when i took 40 mpg + 600 mg of CoQ10.     

 

I also have experimented with  BioPQQ + MitoQ.    I say a good combo.

I can't notice anything. I'm almost out of my first 30 day bottle. If I had to say one thing was definitively different between then and now, I could say that I tend to crash less mid day. Not sure if I am just healthier, my blood sugar is more stable, or this is the key. I'll stop use for a couple weeks before my next bottle.

I've used it several times in the past hoping it would supercharge me, but basically never felt anything. I recently had a 4 week bout of walking pneumonia that really destroyed my energy levels even after the illness was gone. I dosed 60mg the first day and felt a significant increase in energy but had disturbed sleep that night. I dropped the dose to 40mg and have kept it there now for 2 weeks. I definitely feel a boost in energy and cognition, particularly in the morning.

 

Based on experience and the many anecdotal reports of it working great for some with others feeling no benefit, I think that PQQ might be most valuable to those with mitochondrial damage brought on by viruses, etc. Just a wild hypothesis but my experience has sold me on a supplement that I had previously given up on.
