Would you be willing to divulge your supplier should someone offer to host this buy?
edit: I know a source for EVP-6124, but it's ~$130/10mg, possible cheaper when buying more. Dosage is 0.3-2mg/day.


I have been quoted $34/10mg (not very expensive) for 500mg from a trusted lab, so do you guys want to test agonists for the α7-nicotinic receptor?

Sorry, it was for EVP-6124 yes ;) I really want to try GTS-21, but I'm willing to go with EVP-6124 for now. Don't you think it should at least give us a hint if agonists for the a7 receptor has any future? 500mg was minimum order quantity, so we should at least get a month's supply each. 30mg+ each and no more than 20 participants to not make this more difficult than it needs to be. Personally, I'll probably get 50mg.

Around 4h. From what I gather.

Alright; I suggest you start a new topic with "Group Buy" in the title if you want to take the lead on this one. It'd be nice to have that clearly visible.
I haven't been that scrutinizing, but they seem pretty similar to me. Plus, you might catch more participants with EVP; there are a number of people with schizophrenia on this forum who might be interested in EVP but not in GTS.
My memory sucks, but I think GTS lacked the sensory gating enhancing effects, whereas EVP didn't.

Anyhow; $100 buy-in is reasonable and acceptable. How trusted is the lab? (as in: where is it located?)


The lab is the one NSN uses and is located in China. Everyone OK with this?

Are you sure you don't want to be in charge formergenius? Although I'm a full time student, I will probably step up if nobody else wants to do it. I'd hate to see this group buy not getting anywhere.

$3300/100g , $5500/200G, $7200/300G.

At 100G, that works out to $33 per gram. If we took the max dosage of 2mg, one gram would work out to 500 doses. To me, this is very good. so we need 33 people to buy 3 grams each. Is this doable?


It it were only so. That quote was for GTS-21, which requires a much higher dose (100mg-450mg daily).

Does anyone have any suggestion of how we will be dosing? I suspect not everyone (including me) having sufficient equipment to measure out in the 0.1mg range. What would we dissolve it in?

This is exactly what I wondered as well being a smoker (6 months) with my interest in EVP-6124.

After a bit of research i realized that nicotine being a full agonist desensitizes a7nAChR, so with chronic nicotine exposure the cognitive effects would diminish back to baseline and even below baseline during times of low nicotine blood concentrations. A study i read suggested that nicotinic receptors up-regulate from nicotine cessation beyond those of people who have never smoked, causing withdraw symptoms (I believe it was more relevant to the a4b2 receptor).

So I think the only option is to give the brain a rest from nicotine to allow receptors to up regulate and the apply the a7 agonism to see if those much missed cognitive benefits from nicotine can be found again.

Abstract

This study was designed to determine the safety, tolerability, pharmacokinetics and effects on cognitive function of GTS-21 in healthy, male volunteers. A total of 18 subjects were randomized to GTS-21 (25, 75 and 150 mg) or placebo administered three times daily (first 4 days, once on Day 5) for three, 5-day sessions. GTS-21 was well tolerated up to doses of 450 mg/day, with no clinically significant safety findings. C(max) and the area under the plasma concentration of GTS-21 and the metabolite 4-OH-GTS-21 increased in a dose-related fashion; although considerable intersubject variability occurred, it decreased with continued dosing. GTS-21 showed statistically significant enhancement of three measures of cognitive function (attention, working memory, episodic secondary memory) compared to placebo. A relationship between exposure to GTS-21 and the magnitude of the cognitive response was apparent, with maximal effect approached for doses between 75 and 150 mg three times a day. These data indicate that GTS-21 may represent a novel treatment for dementia.

What I don't understand is why they dosed up to three times a day in the study, when the half life of GTS-21 is 12-24 hours (possibly longer than Modafinil).

What I don't understand is why they dosed up to three times a day in the study, when the half life of GTS-21 is 12-24 hours (possibly longer than Modafinil).

Unfortunately, the half life is from what I understand somewhere in the ballpark of 4h. So, I might have to resort to just dissolving it in water and sipping away throughout the day.

Yes, I read that study. There seems to be some discrepancy with the peripheral effects and cerebral.

First thoughts on the GTS-21: Man this stuff is yellow. Comparable to Riboflavin gold, but more yellowy. Take precautions or you will stain your clothes.

After that... No profound altered state after taking the first dose. Skipped the sublingual route since it seemed to bite the tongue a bit, and all the human dosages test seem to be orally administered anyway.
 

On a completely empty stomach, there was only the slightest bit of uncomfortableness as the capsule dissolved, which seemed to go away in less than 5 minutes. 

 

I've not engaged in any cognition tests as this was just a "let's find out if there are any noticeable mood/body changes" type test. The headache that was there this morning seems to have gone away, but you never can really tell with those things because they come and go so frequently. I seem to be fairly awake and not anxious. 

Anyway, just dropped by to see if anyone else had been involved in testing lately.

I've tried 75mg, 100mg, and 150mg doses.

 

Random notes so far: Feels extremely clean. I generally consume moderate to large quantities of caffeine throughout the day, and have headaches if I do not. This morning, I took 100mg of GTS-21 on an empty stomach, with nothing else. I was alert, awake, and no headache in 15-25 minutes.
 

It does not seem to help me with motivation. I've not found anything yet that stacks with it that will help with motivation either, but I've only tried a few things. I'll post more on combo's as I experiment and verify.

I still haven't set down to evaluate gains in memory and cognitive abilities. My main reason for being here has always been focus and motivation, as I struggle with... well whatever you want to call it (ADHD-PI, anticipatory anhedonia, SCT).

 

Honestly, I hope that a substance like this can get a big break and be a replacement for using/abusing nicotine and caffeine. It feels so much cleaner than both. Unfortunately, the cost + lack of long term or indepth safety profile makes it unlikely to happen anytime soon.

As for positive effects (mostly subjective), I noticed a clarity of thought that allowed me to complete a number of projects at work that I had been avoiding recently. I also had a couple of co-workers that remarked that I seemed to be a in a lighter/better mood, and was more collaborative, than usual.

As far as I know, this just hit the consumer market in a very limited fashion. It's actually a quite reasonable price given its "newness". Give it some time... If enough people like it, it will go down in price like Noopept did.

 

I've actually found PRL-8-53 to be the best combo yet. Haven't yet tried it with Coluracetam.
