Hi. I got the Life Extension's NR 60 caps, 250mg per dose. i dosed 2 times daily. and now i finished the whole bottle and i cannot think of a single change with this thing. I usually ride bike a lot but i didnt notice any athletic boost. in fact, i actually got a cold by the time i finished the bottle and i remember last few days feeling really tired and i dont generally feel tired riding my bike. i think arginine was much more helpful. so why is that NR is so praised? maybe LE has crappy fake NR?? is it possible? i kept 1 pill left so i can test it and see if its fake. can you guys recommend some place, a lab, that can test for NR content? I am considering sending it back to Niagen manufacturer so they can tell me best if its real or not. I keep thinking its fake, but maybe it is real and its just that i dont feel it because im different? i dunno.... i really am pissed off at the price i paid so i have to get to the bottom of this.

Life Extension and Thorne use Chromadex branded NR called "Niagen". No need to test it, it is genuine NR hydrochloride.

 

The reason you didn´t feel an athletic boost is probably because NR never gave an athletic boost in human clinical trials. NR didn´t increase VO2max or fat oxidation in human trials. It doesn´t seem to do much to the skeletal muscle at all. It didn´t increase NAD in human muscles and actually downregulated some bioenergetic genes related to the mitochondria in muscle tissue.

NR, like Nicotinamide+Ribose (N+R) and Nicotinamide alone, raise NAD+, and NAD+ induces mitochondrial fission and reduces mitochondrial mass. This is fine if you do it intermittently as it helps eliminate defective mtDNA. But do it continuously and you will not have as much energy.

 

The NAD+ precursor nicotinamide riboside decreases exercise performance in rats

 

RESULTS: The nicotinamide riboside group showed a tendency towards worse physical performance by 35 % compared to the control group at the final 10 % load (94 ± 53 s for the nicotinamide riboside group and 145 ± 59 s for the control group; P = 0.071).

Nicotinamide (NAM) treatment causes a decrease in mitochondrial respiration and reactive oxygen species production in primary human fibroblasts and extends their replicative lifespan. In the current study, it is reported that NAM treatment induces a decrease in mitochondrial mass and an increase in membrane potential (DeltaPsim) by accelerating autophagic degradation of mitochondria. 

 

 

 
The reduction of mito mass can be quite dramatic. See Fig. 1A in the second paper.
 
Bottom line, you don't need NR to do this. N+R is much cheaper and faster acting. High NAD+ levels drive mito fission, which promotes autophagy of defective mitochondria (defective mtDNA, actually). You can override fission with a fusion agent such as stearic acid (10 grams of food grade cooked into brownies) or 50 mg sulforaphane. The first takes about 3 hours and the second takes an hour or less--about the same time for N+R to increase NAD+. In a mito fusion state, you will have more energy.

Chris Masterjohn has some cautions on NR here:

 

 

Apparently, while cycling low doses may have benefits, more isn't better and too much NR can effect methylation.  

 

Once again we find mega-dosing madness is often counterproductive.

NR is like the icing on the cake to already healthy individuals. But that is true of probably 99% of the stuff we discuss here.

March 25th, 2019 on Peter Attia's The Drive. Not an exact quote. Do you take N.R.? Masterjohn. A: If it helps, I took 75 mg with Breakfast and 75 mg with Lunch. I might go up. (He might have changed to not taking it by now)

 

It is an excellent over 2 hour interview. Learned a lot about Creatine and Choline, and various issues such as NAFLD, and Methylation. My point is that these guys change. His video posted in #4 above is "anecdotal" He comments about it at the end of the Attia interview. (Another example of changes is that Attia stopped Metformin due to recent studies. (He still liberally prescribes it for B.G. issues.)

How old are you?  i've noticed is that people in their 30's get very little benefit from NR/NMN.  Whereas those in their 50s+ usually get some benefit, at least as far as conditioning.

linux, im so gullible. i read so much hype about NR and fall for it. but come on, cant blame me. its easy to fall into this trap with all the hype out there.

 

and whats N+R?

i already take broccomax which contains sulforaphane, how do i know its working? any signs?

btw i probably consume grams of stearic acid without even realizing since i take like tons of supplements containing it lol

 

anyway guys thanks for the information on this. i was confused as to how NR benefits someone in what way exactly. it is said to be an anti aging agent, but what should one be experiencing then? and yes, im in my 30s, probably wouldnt be noticeable at all, and if anything, probably cause side effects.

 

but still, were you guys ever curious if your supplement contains the exact ingredient on the label at its exact dosage? in such case, someone must have an idea, must know some place to send for testing.  i hope someone can help me with some information on this. i have been on the LE case for a while as i tried other supplements from them which seemed to work when used by other companies, but not them. so im probably going to test something from them eventually.

Of course, 500 mg of NR with 100 mg of pterostilbine did give the 8% walking boost after the 8th week of the Elysium trial for healthy people 55 to 79. But then again, you know this.

8% for 8 weeks for healthy old people. im not sure if many old people are perfectly healthy but ok. that still doesnt sound amazing. and its a sponsored study. if they compared to something else cheaper more natural they might have had the same result.

anyway, im not going back to NR again ever. in 10 years time when its definately proven to be good for you, im very sure there will be many other better anti aging agents
