Hi, this is my first post on this forum, I hope I can bring value to somebody with it. I was directed here by a 2 year old Reddit post in which a user by the name"painkillerrr" asked to no avail if there is a HDAC inhibitor similar to vorinostat with better BBB permeability. Unfortunately I can't respond to him as the Reddit post is archived and his account removed, however I was linked to an old group buy thread. Apparently there's a lot of demand for methods to get it into the brain better. I decided to use what I've learned in my course to create a much more BBB penetrable drug.

 

I came up with a drug called 7-(methoxyamino)-N-phenylheptanamide

 

The only difference is a substitution of the N-hydroxyacetamide moiety with a carboxamide moiety. This change causes the LogP to increases and the TPSA and MW to both decrease, which should make it easily cross the BBB. I ran target prediction through SwissADME and it should more or less act similarly. Would be cool to see this made in real life and see if it actually worked. HDAC inhibitors seem like they have an untapped potential to heal anxiety disorders, I'd love to see more research done on them.

Apparently 5.5% of Vorinostat crosses the BBB. It would be interesting to see how your drug compares. Is there any Vorinostat group buy I can participate in? Do you have any information on how to acquire it? I'm seeking it to treat benzo PAWS (protracted withdrawal), and am willing to go to great lengths to get it.
