Chronic activation of the MTORC1/P3IK pathway is the primary driver of aging.

MTORC1 inhibits autophagy

MTORC1 induces inflammation through activation of the NLRP3 inflammasome

NLRP3 causes the immune system and other organs to age through overactivation and chronic SASP

Inhibition of autophagy leads to increased SASP, increased cellular senescence, loss of stem cell renewal, cancers, and degenerative disease.

MTORC1 inhibition by rapamycin reverses or ameliorates all age related diseases including:

Diabetes, immune senescence, heart disease, ALZ, Parkinson's, MS, atherosclerosis, muscle atrophy, etc.
