I read rapamycin+rifampciin+allantoin combination doubles the lifespan extension, but what about metformin with the same two?

I also read rifampicin enhances metformin but any info on its inreraction with allantoin? Would it be safe to try the metformin+rifampicin+allantoin combination?

"doubles the lifespan extension" in what, mice? Or fruit flies? At what dosages/schedule?

 

rifampicin is not something to play thoughtlessly around with. "Rifampicin is the most powerful known inducer of the hepatic cytochrome P450 enzyme system, including isoenzymes CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP3A4, CYP3A5, and CYP3A7." (wiki) That's in humans. Other species may metabolize it somewhat differently.

There was another study I recall that said rifampicin inhibits autophagy induced by rapamycin and that can't be good for benefits...?

Also to note metformin+rifampicin+allantoin is far more affordable than rapamycin alone.

I'm under the impression that Allantoin was a better option that Metformin, both have similar mechanisms. I'm not sure about bioavailability and absorption of Allantoin in humans though. Not much data on taking it orally, its all on using it topically in comsmetics. It does look like a very safe compound however.

sorry, but where does it say that metformin and allantoin have similar mechanisms? What, both lower serum glucose? Allantoin is known for its "moisturizing and keratolytic effect, increasing the water content of the extracellular matrix" (google),  does metformin?

Yeah sorry bit lazy of me. I've been looking into these substances recently and I've collected some of the information from various longecity threads and papers.
Basically allantoin has a similar mechanism to Metformin, lowering blood glucose. It was also found that rapamycin +Metformin was slightly toxic, that's why they didn't proceed with Metformin for the rest of the experiment. All combinations used allantoin instead.

where is the combo more affordable?

Metformin is 10 cents per pill and rifampicin is 20 cents per pill from where I buy. Metformin can even be free, I recall there is a free prescription program online that gives out free metformin.

Allantoin is widely available as OTC.

Yeah I read the study suggesting natural substitutes of rapamycin and metformin and both, but allantoin+@ is interesting.

To clarify I am not interested in rapamycin. Rap+rif is toxic but rap+rif+allan doubles the benefits, met+rif enhances the effect of metformin but this study doesn't show any metformin+@ pairs so I have no idea what met+rif+allantoin entails. Even if allantoin overlaps with the effects of metformin, it seems like allantoin is the catalyst of the outcome when combined with rif+@.

Any evidences or studies about metformin+allantoin or met+allan+rif? Only study I found is about met+rif as I mentioned, so they must not be toxic when combined. https://www.ncbi.nlm...ubmed/21270793/
