Most people on this forum take Rapamcin in pill form . I recently purchased 500mgs of Rapamycin in powder form an I dont no the best way to take it . I have been just putting a 5mg scoop on my tongue an swallowing it with grapefrut juice . Should I mix the Rapamcin with somthing else first befor taking it ? This stuff is pricey an I want to make sure I am getting the best benefit out of it .

I made a dilution in pure ethanol. Your way works just as good but i'd say it generates much more work to measure every dose taken. I just take my weekly dose with a syringe and take it orally. I diluted it so when I take 0.1ml it takes 1mg, and I'm taking 0.3ml weekly, for years.

It can't be diluted in water, because it doesn't dissolve at all, I think (but I'm not sure), it would also dilute well in vodka.

I also recommend keeping it in powder form in the freezer, when diluted it can be kept in a fridge but I also keep it in a freezer because I make dilutions that last for around a year and I think the recommendation for oral dilutions of rapamycin it's good on the fridge only for around 1 or 2 months.

Yes, on some rare ocasions I'm unable to like on a trip, but it's like 4 skipped weeks in a year, tops.

How old are you may I ask? I'd only take 5mg if I was over 50 and after slowly upping the dose. 5mg generated minor side effects for me (I started when was 28). The classic mouth sores. Recommended doses are between 2 and 5mg per week and the way I interpret this is you up your dose according to age. With grapefruit which increases drug availbility I'd say 5mg is too much for everyone.

I switched from 1/2 ml of 95% ethanol to 190% everclear . I think the everclear maybe a little healthyer . Still mixing it with abut 5 mg of rapamycin powder an dranking grapefute juce about 3 hours before my dose . I have taken it 5 times so for. Feel great !

If I were you I'd drop the grapefruit and lower the dose to 3mg and up 1mg per month until you reach 5mg.

Grepefuit and rapamycin are pretty much untested, it's not recommended to mix the two and if you do you definitely need to lower the dosage.

The case for rapamycin seems compelling.   I'd like to take it if it will benefit me, but I wonder if it would be good for me.  I'm a 67-year-old male, extremely thin, about 5 ft 11 inches, weight 131 pounds.

Rapamycin inhibits mTORC1 and mTORC2.  Doesn't that inhibit growth of tissues?  I'm not able to gain significant muscle or fat no matter what I do.  I'm sure some of the reason is that I can't eat sugar or other high-glycemic-index foods.  If I eat high-glycemic foods, I can tell subjectively that my insulin goes up a lot and then crashes, and I get weak and shaky.  So I must avoid them.  Wouldn't rapamycin make my extremely thin physiology even more extremely thin?

My fasting glucose is 80 mg/dL.  My CRP is listed as "< 0.2 mg/L".  My creatinine is 0.85 mg/dL.  I gather those are things rapamycin is supposed to help, but those things don't need help.  Of course lower creatinine would indicate better kidney function.  I have noticed trouble remembering certain things; it would be nice to have the memory of my youth.

Your labs look stellar for your age so indeed they don't need improvement.

IMHO, at your age the mTOR signal is already rather strong so knocking it down a few notches with rapamycin makes sense. I suggest start with 5mg once a week and see how you feel. IF your next bloodwork results after, say, 3 months show deterioration, drop it.

As to your memory issues, have you done a few cycles of Turnbuckle's AD detox protocol to see if it will help your mental health?

Aribadabar, thanks for your reply.  I didn't know that mTOR would be elevated at my age, but vaguely wondered if it would be.  And thanks for the note about Turnbuckle's AD detox protocol. I'm familiar with his stem cell renewal and mitochondrial protocols, but I never looked into his AD protocol.  I've read a portion of the thread containing his AD protocol and got the clear message that it's well worthwhile trying it and seeing if there a benefit, and repeating it periodically if there is.  All I notice so far is inability to recall a word or a name from time to time, but it's good to nip things in the bud.

Rapamycin is best taken on an empty stomach.

I haven't come across anything saying it is better absorbed if dissolved in ethanol. If anyone has a reference for that, please post.
