Which blood tests to take before supplementing high dose nutrients (Vitamins, minerals)?

I havent taken my high dosed vitamins/minerals in a while. The product I use is LEFs Two per day.

 

Now I was wondering if I should use this opportunity for taking some blood tests of a certain vitamin/mineral (e.g. vitamin b12). I would be only interested in testing those nutrients where taking a high dose supplement would mask a deeper rooted problem. I am not so much interested to find out about a deficiency due to diet, as I will tweak my diet in the upcoming months anyhow plus that should be solved with taking the supplement anyhow.

 

So is there any vitamin/mineral where a high supp would resolve a deficiency without solving the underlying problem? For example beeing an undermethylator etc.
