is modafinil a weak DAT Inhibitor?

so I was reading about modafinil on wiki it said modafinil inhibits DAT(DOPAMINE TRANSPORTER) 51% which is a lot so it increases dopamine. But I came across a comment that said the following: modafinils ability to increase dopamine signalling is weak compared with amphetamine's ability to increase dopamine signalling, however.

what does that mean? Like ADHD Drugs inhibit more than 51%??? How much do smart drugs inhibit DAT ? Like 90%?

It means... that even though it can cause a 51% decrease of DAT, it still only means that a very small percentage of the drug actually attaches to DAT - it attaches to a bunch of other things first, and affects them - there's not much of the drug left to actually affect DAT.

 

Only if you use ludicrous (as in, DANGEROUS!) dosages of Modafinil will you see a 51% decrease of DAT.

 

 

Most smart drugs/nootropics, have a different mode of action than effecting DAT, or even dopamine neurotransmission -systems at all - most of them affect something called Acetylcholine. (most of the racetams, cdp-choline, alpha-gpc, even Omega-3)

 is 300mg a lot? because according to wiki that was the dosage used and they saw an occupancy of 50%.
