I don't know that Niagen is the most important one (although I take it), but this paper should be in that data base:

Abstract
Adult stem cells (SCs) are essential for tissue maintenance and regeneration yet are susceptible to senescence during aging. We demonstrate the importance of the amount of the oxidized form of cellular nicotinamide adenine dinucleotide (NAD(+)) and its effect on mitochondrial activity as a pivotal switch to modulate muscle SC (MuSC) senescence. Treatment with the NAD(+) precursor nicotinamide riboside (NR) induced the mitochondrial unfolded protein response and synthesis of prohibitin proteins, and this rejuvenated MuSCs in aged mice. NR also prevented MuSC senescence in the mdx (C57BL/10ScSn-Dmd(mdx)/J) mouse model of muscular dystrophy. We furthermore demonstrate that NR delays senescence of neural SCs and melanocyte SCs and increases mouse life span. Strategies that conserve cellular NAD(+) may reprogram dysfunctional SCs and improve life span in mammals.

We know calorie restriction and exercise extend life span. Some studies are indicating the benefit is from increasing NAD+. They call it the convergence of life extension benefit is NAD+ increase.

"In general, NAD+ levels increase in mammalian tissues in response to energy/nutrient stresses like exercise (Canto et al., 2009; Canto et al., 2010; Costford et al., 2010), fasting (Canto et al., 2010; Rodgers et al., 2005) or calorie restriction (Chen et al., 2008). Accordingly, SIRT1 activity is enhanced by all these conditions."

life extension in current world means, living longer but more disabled and lonely in a home for elderly. do you really need this? the future is stem cells! renew your body and control the world, not just extend your life, like a retard

We are talking about extend health span when you are still active until 100 without all the age related illnesses. NAD+ precursors give us hope. It just need more time to be validated in real life.

NAD is not going to make you invincible. you are still getting old and much easier to fall down and break your bones or get infections. also, would NAD fix genetical problems, for example having a gene error for getting asthma or cancer of some type? how would NAD change your genes??

Nobody said NAD+ is invisible. It is a major aging pathway. If we block this aging pathway by supplementing Niagen, we will stay healthy longer and have less cancer. It may not fix the genes, it could fix issues related to bad genes. Cockayne Syndrome is a good example.

To the subject, this recently announced database looks also very relevant:

 

"...Identifying drugs that could extend lifespan in model organisms has received considerable interest (29). Our new DrugAge database (http://genomics.senescence.info/drugs/) is a curated database of drugs, compounds and supplements with anti-ageing effects that extend longevity in model organisms. Although another database of candidate geroprotectors exists, called Geroprotectors.org (30), DrugAge provides a more comprehensive and systematic dataset of life-extending drugs and compounds (31)...."

Other ideas: iodine protocol, pregnenolone/DHEA, and MSM + silicon (monomethylsilanetriol or bamboo extract) + vitamin C.
