Maximizing NAD+ Production Without Downregulating NAD+ Consumption

Hello Everyone,

 

After a few years of becoming more involved in other areas of study, I've become fascinated with the latest research into NAD+ and Nicotinamide Riboside. The evidence seems very clear that by supplementing NR animals and humans can produce more NAD+ that can then be used by a vast number of beneficial mechanisms in the cell. Importantly, activating a number of anti-aging genes such as SIRT 1-7 can help reverse many pro-aging mechanisms such as mitochondrial decline, DNA damage, telomere shortening, etc.

 

I've also discovered that there are other compounds that can assist in maximizing NAD+ by multiple mechanisms, including converting precursors in the salvage pathway into NAD+. However, some of these compounds, such as Beta-Lapachone can work by reducing the consumption of NAD+ by mechanisms that need it for anti-aging purposes.

 

So my question is this: how can we maximize the amount of NAD+ in our cells without using other compounds that hinder the consumption of NAD+ by cellular mechanisms that have anti-aging effects?

 

A few ideas are:

 

1) Find ways to allow NR to be better absorbed by cells.

 

2) Find substances that reduce the limiting factors in the salvage and de novo pathways that do not inhibit the consumption of NAD+ by other mechanisms.

 

3) Practice fasting, caloric restriction, or exercise to boost NAD + levels.

 

Of these options, I think number two is our best bet. NR seems to be adequately absorbed into the blood stream already. Also, the majority of ordinary people in this world are not going to fast, exercise, or participate in CR to boost NAD+.

 

So do we know of any substances that can boost NAD+ or the ratio without reducing consumption?

 

My hope is if we can break through the approximately 3 fold increase limit and increase NAD+ to the level we have at birth, extreme anti-aging effects could be the result.

 

My goal is to reverse my aging process and those of my loved ones and friends -- not just slow it down or stall it.

Yes, the compound which seems to boost the NAD/NADH ratio is beta lapachone.  Three weeks ago two chemists I have been working with for almost a year successfully synthesized it.  I had a proton NMR done and it is indeed pure beta lapachone.  We are now making 10 grams. half of which will be available for scientific research purposes for between 1,500 - 2,000 dollars per gram. Right now we are shooting for June 15th to complete and test the batch.

1,500 - 2,000 dollars per gram how is anyone going to be able to afford this

I read that lapachone downregulates PARP in non-cancerous cells. If this is the truth, then lapachone may not be ideal as a NAD booster.
