How much arketamine can you take intranasally and how often for it to have maximum antidepressant effect and neurogenesis? I've grown tolerance to the substance but it still works, but I have to take slightly higher dosages. I read that arketamine is a 5 times less potent NMDA-antagonist than regular ketamine and 8 times less potent dopamine agonist than s-ketamine, which basically means that r-ketamine lacks psychotomimetic effects. How safe is arketamine then in terms of addiction?

HOWever... the most recent research does implicate the metabolite 6-HydroxyNorKetamine as the component of Ketamine causing the most long-term antidepressant effects - through AMPA-agonism.
I would suggest that instead of using R-ketamine that you try and organise a group buy to manufacture 6-HNK, and then use that instead.

if ketamine works through AMPA there are many other things out there who do that too. also for NMDA, you can make a combo and somehow imitate the effect, no?

Considering that:
-2R,6R HNKetamine molecule is more effective than his brother 2S, 6S
- R-Ketamine enantiomer have several time less psychotomimetic effect than RS-Ket
-buying HNK actually is unbelievable expensive
I'm thinking it have sense to use R-Ketamine for treat depression and suicidality, actually.
Waiting for more experienced answers.

Could you give us an example of other ampa agonists? The only ones I found are on wiki and they are just a dozen, most of them being still in research and never tried on humans before so potentially dangerous. Arketamine also affects the alpha-7-nicotinic receptor which could also be responsible for its antidepressant effect

Arketamine also affects the alpha-7-nicotinic receptor which could also be responsible for its antidepressant effect
The Alpha-7-nicotinic antagonism it displays is most likely responsible for the anxiolytic effects though, the lessening of anxiety and the calming effects - we've got a whole thread on how the Alpha-7-receptors control the output from the amygdala, and how such effects could be useful in treating BORDERLINE.
So, not necessarily responsible for the AD-effects.
For another AMPA-agonist, which also works sort-of like a NMDA-antagonist, then look no further than to good old TIANEPTINE. It works a bit more like Lamotrigine in a sense though, as it MODULATES, aka more stabilises, the electrical impulses controlled by both AMPA and NMDA-receptors - it alters the activity between both of them.
I'm guessing you've never tried Tianeptine? I don't believe it's available in your jurisdiction, so that's understandable - it's available from several reliable online vendors though.

yeah but be careful, tianeptine is partly opioid and you will feel it on and off taking it
