How to take TRU Niagen? (Dosage Question - What's Optimal?)

Bottle says: 2 capsules = 250mg. (so each capsule is 125mg)

 

It suggests: Take 2 capsules daily (so they suggest only take 250mg daily) (is that in separate capsules at a separate time or 2 at once?)

 

But isn't 500mg the optimal dose? (meaning take 4 capsules of Niagen, again all at once or 2 in the morning, 2 at night?)

Also for best absorption and intake:

 

Food or no food?

 

Fat or no fat with meals?

 

Day or night?

 

With special drinks? (Quercetine, tea, orange, vitamin c?)

Hi Forever21! I'm not sure what others would recommend on the board here, but if I were you I would go with the recommended dose for a little while at least (perhaps a month or more). Some people are more sensitive than others and I've heard reports of people getting a tad too much energy out of it to begin with which sort of turns them off to it. If you find that 250mg is too much, just take 1 pill per day instead (125mg). As you get comfortable with it you can try experimenting with some higher dosages. Note that Dr. Brenner now takes 500mg if I'm not mistaken. Keep in mind he's been taking it for quite some time now. I've been taking it for a long time and have experimented with extremely high doses with no ill effects but I believe my body is somewhat less sensitive than others. Everyone's body will be somewhat different despite the fact that all of us have NAD metabolism in every cell. I would also recommend taking it in the morning. I know that when I first began ramping up my dosage, I began by taking another 250mg at dusk. That kept me awake and I had difficulty falling asleep. This no longer happens to me, I sleep like a baby but it was quite irritating initially. I couldn't take it any later than 3pm or 4pm (I typically go to bed around 10pm +/- 1 hr). I didn't notice any differences with regard to taking it with or without food, so I'll leave that to others to discuss if they've noticed anything. You might be able to find some information on that by pouring through the personal experiences thread on NR, but it is quite long.

Also keep in mind, you are fairly young so you may not notice anything consciously even if it is helping you by repletion of NAD within your cells.

 

And if you are inclined or bored out of your mind I would also recommend doing searches on google scholar for anything related to NAD, NR, NMN, NRK1 & NRK2, NMNAT1 & NMNAT2, NAMPT, sirtuins (SIRT1-7 in humans and SIR2 in yeast), PARP (1-16), cADPR, ATP, ADP, CD38, CD157, ENTs, CD73, CLOCK, BMAL1, FOXO1, PGC-1α, mTORC1, CaMKK, AMPK, am I forgetting anything? I think the safest answer to your questions is "no one really knows". Unfortunately. There is still much research to be done, and we are all (not) waiting but still trying on our own. The good news is the forum is full of us trying it every which way.

Jesus what are you? A NAD thinktank? Thank you so much. I'll research.

 

However concerned because I was away from this community for a long time. During that time, C60 came to the scene and then died in this community. I'm concerned that NAD will be the same.

 

All I know right now is that NAD helps/makes/keeps you younger? Sorry, layman here. So I just buy, drink and that's it.

 

But I will research and contribute to the discussion moving forward. Thanks again.

Not necessarily, it's just an interest :)

 

I wasn't around for the C60 discussions but in briefly glancing at that some time ago, it appeared that all of the discussions seemed to center around one study that was never replicated (someone correct me if I am wrong). I have a small non-comprehensive collection of studies around NAD and directly related areas that is well in excess of one hundred at this point. I believe there is a lot more interest from the scientific community around NAD. NAD is critical to human health (we wouldn't be alive without it), but I don't think we can jump out there right now and say that NAD repletion through some of these more advanced precursors helps/makes/keeps you younger. NAD repletion certainly seems to be a viable avenue of research and what research has come out regarding that seems to be positive. I know of only one study that came out some time ago that showed a negative result (https://jissn.biomed...2970-016-0143-x) and no other study that I am aware of has been able to replicate that negative result.  Many of us suspect that NAD repletion through NR or NMN (or both) will improve our healthspan and some of us are hopeful that it will at least modestly improve our lifespan (or prevent/ameliorate illnesses that will eventually take their toll upon us). Many studies promote this idea! It would appear that the safety profile is excellent, so you should feel safe trying it out at this point. If you notice anything negative or positive in your own experience over time, post it over in the personal experiences thread - I'm sure everyone will be interested to hear!

 

EDIT: A lot of people here are very knowledgeable in the topic, so collectively a thinktank perhaps. Several of the members have excellent depth of knowledge and I keenly watch everything they say! New studies are frequently discussed with enthusiasm and if you have any questions whatsoever, post them and I'm sure you'll get some really great responses.

Are you aware of any supplements and/or nootropics and/or drugs (I'm on Effexor/Venlafaxine, an SNRI) or regular Ibuprofen and acetaminophen that NR should not be taken with? 

 

How about supplements it works synergistically with or improves its absorption (Bioperine, vitamin c, etc)
