Has anyone with bipolar tried this? 

 

Tofisopam is an anxiolytic PDE inhibitor rather than GABA/BZD ligand, making it antipsychotic. Able to regulate/lower AMPA signaling. Both dopamine sensitizing in small doses and neuroleptic-like effects in high doses.

 

Does anyone know if PDE inhibition is able to affect serotonin indirectly? It may be ok if just a little but this is important since I don't want to experience serotonin toxicity again.

 

I am looking to eliminate Seroquel XR (quetiapine) 900mg daily, an atypical AP. I know that Tofisopam inhibits CYP3A4 that metabolises quetiapine into norquetiapine... but theoretically couldn't I just reduce the dose of quetiapine to compensate? 

 

I don't actually want the active metabolite norquetiapine and would actually prefer just the quetiapine (considering the pharmacology and my own biochemistry). But how is it eliminated at all? Is it just slower?

 

If the Tofisopam were able to substitute for the Quetiapine, then I would finish with it after some crossover. Can this be manageable theoretically? 

 

I would of course be under the supervision of my psychiatrist.

Have you asked your psychiatrist? If so, what did he say?

Is it possible to get a prescription of it in Australia? I thought it was only sold in some East European countries?

I am not bipolar and do not really have any anxiety, tried it for a couple of weeks. I did not feel any psychoactive effect from it, but then again I am non-responder to most substances.

 

I can not really asses its possible use as an antipsychotic, but it is possible that the PDE inhibition may produce some effect. If it is as strong Seroquel, I do not know, but I would guess not.

Hi bro, 

 

i am using Tofisopam also. 2x 150mg

I am diagnosed with schizophrenia, though i am not 100% about the diagnosis. It could also be bipolar or schizoaffective. But it doesn't matter since the treatment and affected brainregions by the diseases seem to be similar. Never had hallucinations. Had delusions, disordered thinking and mania about 10 years ago and taking different medications since then. Seroquel, Abilify, Amisulpride. Antidepressants i have used: Escitalpram, Moclobemide. 

First time i took the Tofisopam i recognized i adapt more to my surrounding and i am more vigilant. Nowadays i would say, it just makes me more rational. Everytime i take it i recognize a shift in auditory filtering. My perception changes and sound becomes, i would say more "3D" spacial. Music is enhanced also. All these effects it also has taken with amisuplride. Though when taking it withouht the amisulpride Tofisopam is also activating. It doesn't feel like such a huge burden to complete daily tasks. Cleaning appartment, going to work, set a goal. 

I will keep on experimenting with Tofisopam without the amisulpride (so Tofisopam only) and i will check about the combination of Tofisopam+Rasagiline+Pramipexole and see how i react to this. Maybe i get hallucinations then.

Negative sites of Tofisopam: Its an indirect observation, but i think i had little problems with my memory/attention latley. Had a situation yesterday: A girl walking away from me down the hall, 30 seconds letter she reapperas through the stairway from downstairs right next to me. Major dejavu. Also forgetting my PIN code for bankaccount and this happens everytime i start to take the Tofisopam. Forgetting my wallet. And so on. 

Have used Tofisopam in the highest prescribed doses and haven't felt any side effects when going of it from one day to another.
