Hello,

I'm new here. I would love some feedback on my supplement regimen. I put together my regimen for general health, to help with some psychological issues and to prevent conditions that are common in my family history (cancer, Alzheimer's).

I received results back from genetic testing that showed that I was at a significantly higher risk of getting Alzheimer's. I have one of the dreaded APOE4 alleles, having two is much worse, but one is still worrying. So, I would appreciate any supplement or diet suggestions for a long term Alzheimer's prevention strategy. I am 28 so I hope they find a cure before I need it, but I figure it might not hurt to start prevention early.

The genetic testing also indicated that I am a undermethylator (65% of normal). I am also a recovering alcoholic with problems with depression and social anxiety. So any supplement suggestions for these conditions would be welcome.

I eat a low-ish carb, low omega-6 and low fructose diet, with lots of veggies, grass-fed meat, fermented dairy and occasionally some grains. I use CRON-meter and my only deficiencies seem to be Vitamin D (corrected with supplements) and Vitamin E

AM:

Prescribed:
Bupropion XL (depression)
Acamprosate Calcium (alcoholism)

Vitamin D3, 2000IU
Vitamin K2, menaquinone-7, 100mcg
Magnesium Malate, 150mg
Fish Oil, Jarrow Max DHA, 600mg
Milk Thistle, 150mg

For undermethylation:

Methylcobalamin, 1000mcg/2x a week
L-Methylfolate, 1000mcg
TMG, 500mg

PM:

Bacopa, 225mg
GABA, 500mg, occasionally

Some questions:

- I noticed immediate positive mood changes after adding TMG and Methylfolate. Would you recommend adding anything else to correct my undermethylation issue? CDP Choline perhaps?

- I know my diet is low on Vitamin E, does it make sense to supplement mixed tocopherols at a low dose?

- So far the only supplement I take to specifically ward off Alzheimer's is Bacopa. Does it make sense to add something else, like Curcumin, Alpha GPC or even Sulbutiamine? Or is it really too soon to start "worrying" about Alzheimer's? Basically, what would you do/take if you found out you had an APOE4 allele?

Thanks so much for any feedback you can offer!

Your routine looks pretty good overall. Are you aware of the increased cancer risk of that much folic acid (L-Methylfolate) and B12(Methylcobalamin)?

Since when does folic acid = L-methylfolate. I'd like to see studies showing the same with the methyl form. There are some reports of folic acid encouraging cancer growth but I'm not sure about the b12. Please show more studies of this.

Sam-e also helps with undermethylation but be careful with this and start low to test how your body reacts to this.

I did post the study which showed increase cancer risk for folic acid and b12. I'm not saying don't take it, but I did want to give you pointers to the info so you can decide for yourself.

Folic acid and L-methylfolate are different forms of the same vitamin. Do you have any reason to believe methylfolate does not increase cancer like folic acid? Again the decision is yours, I just wanted to make you aware of the risks.

You also might want to look into 1 mg/day Deprenyl.
Read http://www.lmreview....oseph-knoll-md/ for starters and then do some research yourself. If you don't get any apparent side effects from 1 mg/day this might be worth doing, I'm considering it myself although for other reasons.

EDIT: Didn't see that you're taking Bupropion, this might be an issue since Deprenyl has some really bad interactions with other drugs. You might want to look into Deprenyl for sometime in the future when you're taking less meds though. Or perhaps look into switching to Emsam patches for your depression. Just suggestions, your call.

My first instinct is to say that 28 is way too young to be seriously worried about AD. But to be honest, I don't know when the earliest manifestations of neurotoxicity would start to manifest in the different genetic etiologies.

You may want to start reading about neuroprotection and neurogenesis in general, so you'll have a basic understanding of the mechanisms and can follow current research better. I would hop on curcumin (with piperine), but honestly I haven't reviewed it thoroughly enough to say if there would be any drawbacks to starting it at our age. Lion's mane has been shown to protect against amyloid-β neurotoxicity in a few studies, too.
