Niacinamide as nootropic??

I was browsing and found this

Abstract
It is known that endogenic nicotinamide has a tranquilizing and stress-protective activity. The present investigations show the nootropic effect of this drug and its analogs nicomorpholine and acethylnicotinate on acute models of hypoxia and amnesia. The present results revealed that the observed nootropic activity of nicotinamide and its analogs is more expressed than this of piracetam, pyritinol and meclofenoxate. Having in mind the similarity of pharmacological effects of piracetam and nicotinamide (antihypoxic, antiamnestic and anxiolytic) we try if these drugs have electronic-structure similarities. The analysis revealed some similarity of these drugs' molecules in relation to the composition and distribution of polar centres pi- and p-electronic areas) distance between them, topography of separate molecule parts.

How can niacinamide be more powerfull than piracetam? Smells like bull. What do you guys think.

I'm just starting to look into niacinamide myself, and it looks promising.

The reason I'm interested in this is to reverse autophosphorylated tau formation related to early stage Alzheimer's disease (indeed, a safety trial into its effectiveness is currently being conducted: http://clinicaltrial...how/NCT00580931). Niacinamide could be neuroprotective as this kind of tau formation occurs with age, stress, and some chemical addictions.

It appears to have a complicated relationship with resveratrol, as they might both act on SIRT-1 (see aforelinked thread).

I'd be interested to hear from other ImmInsters who have read more about this, and could provide advise as to its effectiveness/dangers.

I took 500mg of Nicotinamide today, and noticed effects within about a half hour. My response was a fairly strange combination of increased concentration and alertness combined with anxiolysis and a sort of calm, sluggish physical feeling. Reminded me a lot of benzos, except being with clear nootropic action. I'd combine with stimulants, as I personally like being a little speedier; but it looks like a great alternative to benzos for anxiety.

Also an effective anti-acne medication (I've tried topically, supposedly orally also) and in high concentrations a safe skin bleaching agent.

I was on 500mg/d for 10 months. It did relieve some anxiety. I didn't experience any nootropic effects and it didn't do anything for my acne. Stuffs really cheap. Like $10 for 300 day supply. Give it a try.

I tried 500mg of Niacinamide with 400mg of Caffeine last night. Helped clear up a lot of the sluggishness. Definitely a nootropic, albeit not an extremely powerful one. This stuff really works as a topical acne medication at 4% concentration, as in fully cleared up my acne within a few days. Previously I'd been using bondo hardener (which contains 30+% benzoyl peroxide, Proactiv is weaksauce), and this is more effective.

Had some Niacinamide lying around, saw this thread and started dosing at 3g/day acutely then down to 1-2g/day. After a few days of this with zinc and vit C and magnesium I feel pretty lucid when I am going about my day. It's hitting an aspect of brain fog I had long neglected. I am of those people who need niacin more than others hence I'd be deriving a lot of benefit from supplemental niacin. Many thanks for the recommendation. I should have started this way back when I learnt about Abram Hoffer's work in curing many conditions with niacin.


Plain niacin is amazing too! It's much cleaner on the body once you get used to the flush and it's possible to dose that heavy, which can be very rejuvenating.

It's actually the other way round!

Niacinamide is slowly metabolised and taxing on the liver, whereas niacin is in and out smooth as can be. Abram Hoffer had his patients between 3-6g/day I believe, maybe even more! And this was kept on long term, some/most of these patients administered Niacin high dose for the rest of their lives to manage their conditions. Niacin is very safe.


The flush reaction is nothing. Stop being a pussy, it's a little flush... Yes, you go red, hot, itchy for a little while. So what? You're not going to die of a niacin flush... (inscribe that on my grave stone if I ever die of Niacin OD :-D ) Many bodybuilders pride themselves with such boisterous ego yet shake at the idea of experiencing a niacin flush, the mind boggles.

I´ve had also relative good effects. I have the assumption that Nicotinamide has some effects on glucocorticoids by interacting with 11 beta-hydroxysteroid dehydrogenase, which is the degradator of Glucocorticoids,

but I´m unsure if definitively and to what extend.

Niacinamide is a mast cell stabilizer, whereas niacin induces mast cell prostaglandin release (though the net effect is supposed to be reduction of mast cell activity in the long run). So if anything Niacinamide will lower pathological histamine levels. Figured I have mast cell problems after I started getting inexplicable rashes all over my lower legs and hands. Can't tolerate even 50 mg of niacin, but up to 1.5 g of Niacinamide daily is great. First effective nootropic I tried.
