Hey. I've been reading about the dosing for phenibut and picamilon and something seems odd. Phenibut's dosage is about 750mg a dose but Picamilon is about 70mg a dose. My question is, why such a big difference in dosage. Isn't Picamilon just a GABA with a niacin attached and Phenibut is just a GABA with a phenyl group attached? So why does it take less Picamilon to get the anxiolytic effect?

Phenibut is effective at doses as low as 150mg in certain individuals. It has alot to do with persoanl biochemistry to be honest.

Biochemistry isn't as simple as niacin group vs phenyl group.
You also need to take into account:
- bioavailability: how much of the substance gets absorbed in the GI tract;
- XlogP3 value: how readily does the substance cross the blood-brain barrier;
- which receptor(s) does the substance bind to;
- what is the binding affinity of the substance;
- half-life of the substance, at what rate is the drug metabolised.
You also need to take into account:

Once broken down and inside the BBB both substances have the same GABA neurotransmitter, so wouldn't half-life, metabolism, oral bioavailability, and binding receptors be the same? I'm just interested in the GABA part, I know that niacin and the phenyl group act differently once they are broken apart from GABA.

Ok I found out more.
Phenibut is a GABAB receptor ligands antagonist
Picamilon is GABA itself.
So they indeed do different things inside the body.
The question now is, can I take both a the same time to get more of an anxiolytic effect?

You totally can, I don't think it would have any serious toxic reactions at normal doses. There might be some sedation. You may be asking for a very long nap combining the two, or you might not even notice a difference at all.

Picamilon is a joke to me. Is there really any evidence that it actually crosses the BBB and then drops off the Gaba molecule into the receptor sites? I don't know, but I know I felt practically nothing from it. Looks like I'm an InternationalAntiAgingSystems propaganda victim for buying it. (though I applaud anyone who has had success with it!0
