The choice between carnosine or beta alanine is very frustrating and I'm surprised their is no clear consensus, the price of carnosine is not justifiable but the beta alanine studies are on muscle carnosine levels and don't refer to glycation

I actually haven't seen that article until now. Looks like an interesting discovery. It could be helpful to other people.

2. My significant other who has a family history of depression has benefited significantly from 500 mg/day L-carnosine (AOR's).

Hey everyone! I want to revive the thread. I've been on carnosine for the last 10 and a half months or so. For the majority of that time I been using extremely high doses. 4.5 grams a day to be exact. Carnosine is one of the main supplements that I take. I also take taurine, krill oil, and Acetyl L-carnitine. I have seen great effects and was wondering if there is anyone new to the supplement lately. I'd like to talk more about it

I take 1 gram a day...starting ~2 weeks ago. I just got it because it was on sale.
I haven't noticed anything I attribute to it.
I do feel slightly more 'awake', but timing makes me attribute this to Seanol, which I started at roughly the same time (though not exactly the same time).
Will update (for either) if it warrants merit in a few weeks.

It's more of a longer term supplement. You have to really be on it a while to see the results. I am continuing to slowly see results in a glacial fashion. Although I do take larger doses than most people my muscles have been getting progressively stronger. It seems as though my fast twitch muscles are working in over drive or something. When I was in school I used to play baseball and have several bats at home still after not having played for a few years. A few times a year I go out and swing and play with a few friends. I have noticed that the bat feels slightly lighter but more interesting is that when I actually swing its so much faster than usual.
Also, i have noticed that my tolerability for fatigue for low to moderate intensity exercise is limitless. For high intensity exercise it seems the same as it always was and I tire as I normally have in the past.

edit: i can see it now. it was the exercise part. i agree, my (poor) tolerance to exercise has improved, pretty much overnight.

but, to add to this discussion, i'm getting unexpected results from carnosine supplementation (2x500mg): more energy and motivation, clear mind, less anxiety, less appetite, more talkative, better exercise, etc.
the effects are pretty immediate, start 1-2 hrs after taking the pill and last about 6 hrs. there is also some dopamine like stimulation and warmth in the face. the effects are much stronger when combined with inositol and i'm thinking they are both synergistic in terms 5HT2a antagonism and 5HT2c agonism. i don't understand why though.
maybe it's the antidepressive effect of beta-alanine?

I disagree. I'm taking carnosine, but not carnitine and the energy boost is unbelievable and no tolerance at 1.5 weeks. Another family member started taking it too and reported the same thing. this is very surprising and not placebo as i don't experience any placebo effects with supplements) and i didn't expect such results after reading about it. I only expected decreased glycation and telomerase effects that i assumed would not be subjectively felt one way or another.
i did some googling last night and one study suggested carnosine stimulating H3 receptors. that would explain the stimulating effects.
as for the skin part. i took a pre picture of my wrinkled face and will do a post picture after 2 weeks on it.

Carnosine has been studied extensively in Russia during the Space race. Roscosmos scientists identified l-carnosine as one of the best supplements to combat accelerated aging of the cosmonauts in the space. However, on its own, they found, l-carnosine is not that effective, as only minuscule amounts reach mitochondria where it does its magic and its effects last for only ~2 hours after intake. Combined with other synergistic substances it shows itself in full strength even at much lower doses and is effective for as long as 12 hours after oral intake. As OP found himself, it takes about 2-3 months for l-carnosine concentration in mitochondria to build up and to feel its full potential.
Personally, I've been taking one l-carnosine formula* from Europe for over two years now, 1x125mg 12 hours apart (total 250mg daily) on an empty stomach. Combined with hydrogen rich water it is, in my opinion, the best antiaging supplement out there. It makes muscles and hearth stronger, brain faster, and every cell younger; like 10 years younger. It does take 2-3 months till you feel these effects but it's worthwhile.
* the formula contains 125mg l-carnosine and 20mg l-carnitine, among other things

Do we know the time frame in which this histamine receptor stimulation takes place? In other words, is carnosine supplementation in the morning a problem if one suffers from chronic insomnia?

I did some more experiments and took low dose (50 mg) of niacin either before or after carnosine (not on the same day though). the post dose had a whole lot more flushing, that seemed to support the histamine theory, until i realized niacin flushing was from PGD2 and 5HT rather than histamine: http://jpet.aspetjou...ntent/327/3/665

according to one of the (then) research associates of the institute and now almost retired mitochondria medicine specialist. He has his line of l-carnosine product (quite effective & somewhat pricy) in Europe. He personally examines you via HRV and proposes the frequency and dosage. The majority of his customers are professional athletes, execs in their 60-s, and kids with autistic spectrum (google karnozin extra, if interested).

Do you have the reference to the vitamin E claim? Because I think that VECAR (a novel heterodimer of α-tocopherol (vitamin E) and carnosine) is not the same as taking Carnosine and vitamin E separately and, in fact, what I've found is
Dietary vitamin E, but not carnosine, increased levels of vitamin E
Muscle vitamin E concentrations were not significantly affected by dietary carnosine.
Carnosine supplementation significantly increased E levels in liver and heart, but not in skeletal muscle, serum, or erythrocytes.
but I'm not able to find any research about Vitamin E increasing carnosine efficacy.

The results are in re: wrinkles. I have pre and post (3 weeks of carnosine) pictures of my face and there are more wrinkles after! So much for that.
I can't think of a biochemical reason why carnosine would be expected to reduce wrinkles in 3 weeks. At most, it might slow the accumulation of wrinkles slightly by slowing glycation. You'll still have to wear sunscreen though, as carnosine isn't going to stop the UV damage that is far and away the biggest cause of wrinkles. Finally, if you take pictures every month for a year, you will probably find the appearance of your wrinkles varies over time depending on your weight, hydration status, moisturizer use and the current relative humidity.
