Mitochondria are important in aging, and the process by which they become damaged and contribute to degenerative aging starts with the fact that they emit reactive oxidants as a byproduct of their normal operation...

 

...the most potent anti-oxidants were those that targeted the...mitochondria...the most potent mitochondrial targeted anti-oxidant was Tiron...which provided 100%, protection of the skin cell against UVA sun damage and the release of damaging enzymes causing stress-induced damage...

  • Resveratrol...was found to protect against 22% of both the ultraviolet A radiation and stress-induced damage.
  • NAC...offered 20% protection against oxidative stress and 8% against UVA,,,
  • curcumin offered 16% protection against oxidative stress and 8% against UVA.
  • In comparison Tiron offered 100% protection against UVA radiation and 100% protection against oxidative stress...

Tiron chelates released Iron in the mitochondria where the released iron would normally damage mtDNA.
Metals like iron and copper also catalyse the reactions that lead to Advanced Glycation Endproducts and Tiron looks to be effective, in vivo, here too.

It has undergone tox studies in pregnant rats with fetal size issues showing up at 3000 mg/kg/day, making the no observable adverse effect level (NOAEL) for maternal and developmental toxicity; 1500 mg of Tiron per kg per day, in rats.

That works out to a NOAEL of 486 mg/kg/day  in humans, or 3400 mg per day in a 70 kg human.

NB That the above is NOT the recommended dosage, is the highest safe dosage

 

There are no studies I can find that give any indication of the optimal dosage in humans, so taking Nilotinib would be pretty experimental...

Yes its taken orally Ceridwen, but seems to have quite low bioavailability.

Perhaps this can be fixed with lysosomes or Cyclodextrin or something.

In conclusion, in the present study, we have shown the high binding capacity of the pigment from black sesame seeds toward heavy metals, such as lead, cadmium, and mercury, that are commonly found in food. The highest activity is associated with neutral pH values simulating the intestinal environment compared to the acidic pH values peculiar of the gastric compartments. Even at low concentrations (0.05 mg/mL), the pigment proved able to remove the metals to a significant extent (>65% for cadmium and >90% for mercury and lead). At such doses, the pigment is able to remove the metals completely at the highest levels that have been reported in contaminated food. A good chelating ability against lead and to a lower extent cadmium is retained by BSP, even with the metal at concentrations up to 200μM. The binding activity is not lost following digestion as simulated using a model of the gastrointestinal transit, and on the basis of competition experiments, it should not interfere with the absorption of essential metals, a drawback exhibited by several dietary supplements, such as cereal fibers.In combination with our previous observations indicating the marked antioxidant properties of BSP, these results highlight the potential of this low-cost, easily accessible material from plant sources as an ingredient of functional food or as a food supplement.

BSP should be a filler in every supp IMHO.

 

 

There is also intriguing research showing additive and even synergistic effects from combining intra and extracellular chelators.
The intracellular chelator gets the unbound metal out of the cell and 'passes the ball' to the stronger extracellular chelator for excretion.

 

The combination of a weak chelator which has a better ability to penetrate ceils, with a stronger chelator that penetrates cells poorly but has a more efficient urinary excretion, may result in a synergistic effect through iron shuttling between the two compounds.

What makes Tiron intriguing is its ability to not only get into cells, but also into the iron rich mitochondria within cells where any unbound iron causes serious damage.

(Tiron gives 100% protection against sunburn due to this  property..! Think 'no skin aging due to sun exposure...')

 

Naturally its important to know the half life of your chelators for efficient chelation therapy...  That and the clinically effective dosage is proving very difficult to ascertain!

 

 

It would seem that the Mitochondria damaged by unbound iron may also overload the lysosome with iron when recycled.
The iron causes a lysosome damaging 'fizz' when the acid within them reacts with it.
I am sure you are aware of the importance of a properly functioning garbage recycling 'production line' within cells...

Hence my getting and wanting to 'test' Tiron.

I take Inositol in the morning due to autophagy being upregulated at night/fasted and it having a half life of around 5.5 hrs in babies. Probably longer in adults.

tiron should receive his Tiron on monday, I will have to wait a little longer.

My research into dosage with Tiron did not turn up much useful information and was quite extensive.
I dont have the time I would like to look further.

Thanks for counting me in for 1 x 25g vial of Tiron.

 

At dosage of 300mg/day a vial lasts 83 days.

This allows for a 1 month trial with up to triple dosage.

I will be testing it for effects, and modulate the dose.
