Tianeptine is a mu-opioid receptor agonist, and delta-opioid receptor agonist to lesser extent. These are responsible for the rapid anti-depressant effect and also possible sedation.
The SSRE-effect is rather weak and doesn't have much to do with its therapeutic action, but modulation of NMDA/AMPA-glutamate function enhances certain cognitive functions.
