So I've got 100mg of Polydatin in front of me in a Pteromax pill. Does anyone know of a source for pure Polydatin?

What got me interested in this was this study:

Their neuroprotective effects on modulation of protein expression of α7 nAChR, α3 nAChR and synaptophysin (SPY) on SH-SY5Y human neuroblastoma cell line (SH-SY5Y) were evaluated by means of Western blotting. Results: Two potential compounds, polydatin (C1) and emodin-8-O-β-d-glucoside (C2), were detected and identified in our study. The biological tests showed that both compounds C1 and C2, respectively, at concentrations of 0.1 and 0.25 mg/mL significantly increased protein expression of α7 and α3 nicotinic acetylcholine receptors (nAChRs) in SH-SY5Y cells. Moreover, C1 and C2 at 0.1 mg/mL significantly reversed the Aβ1-42-induced decrease of α7 and α3 nAChRs protein expression in SH-SY5Y cells. In addition, C2 at 0.1 mg/mL significantly increased protein expression of SPY in SH-SY5Y cells and Aβ1-42-induced SH-SY5Y cells whereas C1 did not provide any positive effects.

This could be good for short term memory as nAChRs are critical to short term memory formation.

Nicotinic α7 receptors enhance NMDA cognitive circuits in dorsolateral prefrontal cortex.

...
Here we explore α7-nAChRlocalization and actions in primate dlPFC and find that they are enriched in glutamate network synapses, where they are essential for dlPFC persistent firing, with permissive effects on NMDA receptor actions. Blockade of α7-nAChRs markedly reduced, whereas low-dose stimulation selectively enhanced, neuronal representations of visual space.

So 45 minutes after taking Petromax (100mg Polydatin, Resveratrol 250mg, Pterostilbene 100mg) and 250mg CDP Choline I got an 8 on paired associates on CambridgeBrainSciences.com which is about as good as I can do. This test is super consistent for me at about a 6 or a 7 on a good day. I'd say this is comparable to the effects I get from galantamine. I wonder what mixing both of them would do?

BTW, I also get this interesting effect when I'm looking at the tiles on paired associates. It's sort of as if when I look at them there's a bit of a glow around the actual object inside the white tile. Like my brain is focusing on the shape and trying to ignore the white space around it.

More on galantamine and nachrs

http://www.ncbi.nlm....pubmed/22932776

These findings suggest that galantamine increases hippocampal IGF2 levels via α7 nAChR activation in mice and imply that the effect may contribute to its neuroprotection or neurogenesis.

Hi Twitterers.. See my earlier post for why Polydatin is an interesting substance from a short-term memory perspective.

Edit.. Wow.. Just hit all my high scores on cambridgebrainsciences.com! Nice combo!

Already ordered some. Looks like they are back-ordered though. I've only taken this once but it sure looks like a winner of a stack so far.

I wonder how this would affect a choline dominant person?

Adding the citation for my first study so I can find it later: http://www.ncbi.nlm....pubmed/24143857

Thanks for sharing this.

Lately I've descended from goldfish to vegetable, so I've ordered PteroMax to give it a whirl. Predominantly because the visual effects might be beneficial to my HPPD, and the NMDA action may be beneficial to my depersonalization. Will report back once I've tried it.

Abelard, how long did the effects last for you approximately? Have you tried 200mg?

Maybe niner can help us here with the pharmakinetics, but I believe the major pathway for polydatin metabolism, is splitting off the sugar moiety from the molecule, leaving.... RESVERATROL! and resveratrol in turn is glucorinated, to a molecule similar to polydatin. Eventually everything is either excreted by the kidneys, or goes back to the liver and gets converted to resveratrol again, i.e. hepatic recurculation....

Anyway, I think the only benefit of polydatin might be to get higher levels of circulating resveratrol than you might get from resveratrol alone. Maybe you can the same effects from resveratrol in an appropriate dose, as you are looking for from polydatin?

OP, are you still getting the same gains from polydatin or have you dropped it?
