And advice, don't get the niagen crap. 150-300 mg is miniscule. I finished two jars in 4 weeks and cost me 100$. One gram a day is the goal and this group buy is the key to that goal :)

Not crap in the sense of quality. Niagen is legit. But crap because the dose is miniscule compared to the study dose. I'm pretty sure also, it's two caps totalling 250mg, not one.

If memory servers me right, weren't Sinclair's mice dosed by injection? Or was it an in vitro study of tissue cultures that started this craze ? Not that I don't think NAR (gotta call nicotinamide riboside something catchy) won't prove beneficial in some ways, but I don't expect miracles from oral supplementation.

I believe that the study you are referring to was using niacin mononucleotide (sp?). NR is it's precursor, and can be taken orally.

But irrespective, oral won't get the serum levels you get with injection.... or in tissue cultures bathed in the stuff.

I have been following this thread since the beginning and I too am interested in being involved 0.5 to 1KG...

My only concerns are , legitimacy/quality of the product, shipping to the UK and storage of the product (does it last and does it degrade over time)

If my concerns are eased I'm in. :)

If I saw correctly, shipping will be about $21.60 for 500g (to Sweden atleast) and $29 for 1kg (again, Sweden) assuming nootropicsdepot uses the same shipping as they have now.

His research used NMN, NR is converted into NMN in the body.

NR is orally available in mouse studies and there's no reason to think humans are different.

"NR-fed mice upon HFD was not due to reduced food intake, as NR-fed mice actually had a tendency to eat more"

"NR treatment dose-dependently increased intracellular NAD+ levels in murine and human cell lines (Figure 1A), with maximal effects at concentrations between 0.5 and 1 mM. In C2C12 myotubes, the Km for NR uptake was 172.3 ± 17.6 μM, with a Vmax of 204.2 ± 20.5 pmol/mg of protein/min. Unlike NA, both NR and another well-described NAD+ precursor, NMN (Revollo et al., 2007), did not activate GPR109A (Figure 1B), hence constituting valuable candidates to increase NAD+ levels without activating GPR109A."

I looked it up. A double nuaght capsule will old up to a gram. I am suspicious of this muner nut run with it. 32 of these throughout the day. Maybe make a set of brownies as snorting would cause serious snot issues.My thought is that the 5 grams of NMN is better. That would be injected though. It is a phophate like SKQ1 but how would that absorb through the digestive tract? Another thought is if you inject 5 grams into your body, how do you know if that is really effecting your entire system? I am sure your blood workup would be effected.

32 grams mean that one key is a month. If they can make pharmacuetical cocaine in China for 5 cents a gram then why are we paying anything serious on all of this stuff? The people at Medicass are still yapping over the SKQ1 costs. It stll looks like a long chain meth to me.

I am sure your blood workup would be effected.

I can't wait to try this stuff. I would love to try it for a few months, then see my doctor, and have his eyes bug out from my numbers all coming back as if I were 20 years younger.Of course, we don't know that would happen. Maybe it won't do anything.

Also, the dosage is confusing me. Some have had side effects (crashes) at small dosages of 250mg/day, even every other day. Others are talking about 32gm/day! Where did the 32gm/day come from?

It came out of dpaxton's behind.

I calculated the amounts required earlier in the thread. At the dosage used in the Niagen formulation, a years supply comes to just under a hundred grams. Some people had rebound effects at that dosage, causing them to stabilise at half the Niagen recommended dose, every other day. Following that protocol works out at 23 grams per year. Unless you guys are planning to acquire a 10-40 year supply, which would degrade in the meantime, you might want to reconsider how much you're throwing in for.

If you read a little closer you'll see that he said 32gm not 32mg... His notation was a little unusual.

If you scroll down they show that 300mg per kg per day was needed to chow NAD+ increase in muscle. I am 85 kg so that is 34 grams. Notice that all tested increased nad levels in the liver but NR and NA worked in the muscle tissues. I wonder how much it costs to feed them that much NMN. I am looking at all of this to make a mix of things for a one a day that is not a hand full of gel caps. Maybe mix in some TA 65 and some others.

You need to convert the dosages to human dosages.
Assuming mice:

300 * (3/37) = 24.3243243243 mg / kg (let's round that to 25)

25*85 = 2125mg = 2.125 g.

Also, how is it going, is anyone looking for a manufacturer still? I think it is impossible for people to say how much they want before they know how much it costs, and as previous comments have mentioned, they need much less for humans.

Well, that's why I'm only interested in a quarter kilo. But it will end up being for me and several family members. I'd rather buy too much than too little, especially when the price is so low relative to the pills found on amazon.com.Besides, it's not like the stuff will degrade over the next few months, will it?
