I see this as a good stopgap medication, but the real reason for PD and related dementias seems to be the disruption of the 'junk recycling' chain by pathogens and/or Iron etc, in recycled misfolded proteins and mitochondria etc, reacting violently with the acids in the lysosomes and damaging them.
ie: the intracellular 'recycling chain' ends up with a broken link in dementias.

Should this be interpreted to mean that if any one of several situations occurs that could end our supply, and I don’t know how likely it is that that could happen, Sinemet and other similar therapies are of no value - we’ve put all our eggs in the nilotinib basket?
It means that if you stop taking N; the PD will proceed again as it did before you started MarkB.
ie: You/the N has to keep removing the misfolded proteins, or it stops working.
Except for bad genes; dementias seem to the result of a breakdown somewhere in the intracellular recycling 'production line'.
Hence my previous postings on the lysosome (the end of said 'production line'), PARKIN and PINK1 etc...
Its pointless upregulating autophagy with N/PARKIN upregulation, if the lysosome has been damaged by the violent reaction between the acids in it and excess iron etc...
ie: Anything that upregulates autophagy (circadian rhythms etc) should be of great interest to everyone here.

Is anyone familiar with the diabetes drug, Liraglutide, as a potential treatment for PD?
It's currently being tested on Alzheimer's Disease patients. Supposedly, it works well in late stage AD.
"When injecting liraglutide once-daily... in two month old mice for 8 months, the main hallmarks of AD were much reduced. Memory formation in object recognition and Morris water maze were normalized and synapse loss and the loss of synaptic plasticity was prevented. In addition, amyloid plaque load, including dense core congophilic plaques, was much reduced. Chronic inflammation (activated microglia) was also reduced in the cortex, and neurogenesis was enhanced in the dentate gyrus. The results demonstrate that liraglutide may protect from progressive neurodegeneration that develops in AD. The drug is currently in clinical trials in patients with AD."

Does anyone know if anyone in the first or second group buy for N research have MSA? If so I'd be very interested in any results you may have from your research. I am involved in a fund raiser to a small clinical trial thru Georgetown University for MSA patients to be treated with N. We have raised over 50k but need 150k more. It would help to know if N has helped with anyone with MSA like it helped with PD. Is there away for a researcher to contact me with any results he might have had? Thanks.

Food/fats increase the effect of N.
The safety info recomends taking it with a 2 hout gap before and after food IIRC.
The fact that food/fats increase feeectivness is being used to get more bang for your buck by a # of researchers.
CYP3A4 inhibitors like piperine and grapefruit juice have the same but more powerfull effect and is also being used.
BHT (Butylated Hydroxytoluene) is a CYP3A4 upregulator that may well be usefull in case of overdose...

I'd like to add a followup to my comment about the the diabetes drug, Liraglutide, as a possible treatment for P.D. From what I've read, Liraglutide and a few similar diabetes drugs look like very promising treatments for both Alzheimer's and Parkinson's. These drugs belong to a class of chemicals called "incretin mimetics" or "glucagon-like peptide-1 (GLP-1)."
As I've learned over the years when researching prescription drugs, you can discover some interesting things about a drug by doing an internet search that includes the name of the drug along with the term "substitute" and another search with the term "supplement."
Well, here's what I found. It seems that there's a supplement that might mimic Liraglutide. There's something called "green-plant membranes" or "thulakoid" which increases the release of GLP-1 in the body. According to the article below, the supplement is used as a diet aid.

There's one brand of the green-plant membranes on Amazon.com called Appethyl. At $30 for a month's supply, it seems reasonably priced. If my mother wasn't taking "N," I might have given her this supplement.

Observation but I've been placing my dosage under my tounge and letting it dissolve and it seems to disappear making me wonder if this is the most effective way to utilize Nilotinib.-source(my own Speculation.)
I don't think there is any research available on sublingual dosing Ark, so there is no way of knowing what effect doing so has.
At a guess it should increase effectiveness, if the molecule is small enough to pass through the membranes in ones mouth.
So what you need to do is find out the molecule size and then the maximum size of molecules that do get in.
IIRC the hydrophilic, hydrophobic, lipophilic etc properties of the molecule also need to be considered.
The information is all here somewhere, but I dont have the time for research I used to have.

Does this work on MS? I don't need it but I know some who do. I wonder if its good for prevention as well as cure? When you get older maybe it will keep you sharp?
