How to take pregnenolone but avoid 5HT2A activation?

It's taken me a long time to figure it out but it's now apparent that pregnenolone has affinity for or some indirect effect on the 5HT2A receptor and that is, among other things, what I am hypersensitive to. Ironically, reduced activity of the NMDA receptor can supposedly increase activity of the 5HT2A receptor. Perhaps it's both excessive and insufficient NMDA activity that can cause activity at this receptor, explaining both schizophrenia and autism (hypofunction and hyperfunction).

 

Anyway, I'm very sensitive to 5HT2A activation. I don't like inositol, I find that it has paradoxical effects. I've recently discovered that BCAAs, or at least l-leucine, cause insomnia and poor sleep quality, even as little as 100mg or below can do this to me.

 

I want the benefits of pregnenolone on cognitive health without the psychiatric disturbances from its serotonergic effect.

have you even tried pregnenolone? ive tried two versions so far up to 100mg both micronized and normal oral pills and i cant seem to feel anything at all. the serotonic effect would be a nice addition ill think but sadly it doesnt even do that in prolonged use. not sure why there are so many positive studies associated with pregnenolone when it doesnt seem to be bioavailable in its normally taken state, pills. i guess i should look deeper into the studies perhaps they are using patents and various forms of non-comercial pregnenolone sources that are probably injected or snorted

Yes, I've obviously taken pregnenolone, 50mg daily for a prolonged period. I've tried 10mg, 15, 25, 100, etc. Even 10mg has serotonergic side-effects.

You can obviously feel pregnenolone, if you can't then you've been buying some fake stuff. At 50mg it feels like adrenergic receptors are affected or something, you may feel oddly dizzy or spaced out or something, difficult to explain. 100mg tends to give razing heart and anxiety.

 

Maybe it's a matter of the differences between pregnenolone sulfate and just regular pregnenolone?

It's really bizarre because some pregnenolone versions (i.e. Swanson's) feel just serotonergic and mildly stimulating while others, e.g., the above seller, feels strongly stimulating but more mildly serotonergic.

You didn't understand any of my points. How is what I said only negative and why would it matter if it was negative? I don't understand the nature of your comment.

 

As for the point of using pregnenolone, it is to get all the cognitive-boosting effects.

thats what i meant, reason for using it. but as i said before, the studies show pregnenolone is various non commercial types has been shown pro-cognitive effects and i dont think the cheap type sold to the masses will do much. i tried to get allopregnenolone a while ago,but the price was so high and nobody else wanted to do group buy so i kind of gave up on pregnenolone since

It might be interesting to get a Pregnenolone blood test and see what your blood levels are.

 

"Life Extension suggests that serum pregnenolone levels of 180 nanograms per deciliter (ng/dL) for men and 200 ng/dL for women may be optimal."
 

I got a "Pregnenolone, MS" test as part of a panel in 2013 that showed 75 ng/dL. So, I guess I'm severely low. But, the reference range for the test only gave an upper limit of <151 ng/dL, as if anything below that is fine. But that upper limit is below what LEF says is optimal. So, I don't get it.

How on Earth do I ship my blood? I surely can't get my doctors to do it. I live in a country with socialist healthcare so we don't pay for the medical services at all. But sadly we can't get things tested, they are very strict about testing things.

According to Wikipedia sources, pregnenolone readily converts to allopregnenolone and pregnenolone itself is dirt cheap like cholesterol powder. So you should be able to cheaply buy bulk pregnenolone and take 100-200mg a day, which would drastically increase allopregnenolone. The only issue I see is the annoying pharmacology of pregnenolone and customs issues because it would look a bit suspicious if you bought raw pregnenolone, I guess.

 

 

Oral administration of 50 or 100 mg pregnenolone has been found to have minimal or negligible effect on urinary levels of testosterone and testosterone metabolites, including of androsterone, etiocholanolone, 5β-androstanediol, androstadienol, and androstenol (and/or their conjugates), and this suggests that only a small amount of pregnenolone is converted into testosterone.[17][18] This is in accordance with findings on the conversion of DHEA into testosterone, in which only 1.5% of an oral dose of DHEA was found to be converted into testosterone.[17] In contrast to the androstanes, 50 or 100 mg oral pregnenolone has been found to significantly and in fact "strongly" increase urinary levels of the progesterone metabolites pregnanediol and pregnanolone (and/or their conjugates), whereas pregnanetriol was unaffected.[17][18] Unlike the case of oral administration, transdermal administration of 30 mg/day pregnenolone cream has not been found to affect urinary levels of metabolites of any other steroids, including of progesterone.[18]

Sripada et al. reported that oral pregnenolone is preferentially metabolized into the neurosteroid allopregnanolone rather than into other steroids such as DHEA or cortisol.[19] In further research by their group, a single 400 mg dose of oral pregnenolone at 3 hours post-administration was found to result in a 3-fold elevation in serum levels of pregnenolone and a 7-fold increase in allopregnanolone levels.[19] Pregnanolone levels increased by approximately 60% while DHEA levels decreased non-significantly by approximately 5% and cortisol levels were not affected.[19] Another study found that allopregnanolone levels were increased by 3-fold at 2 hours post-administration following a single 400 mg oral dose of pregnenolone.[19]
